Calibrate the System

Before you calibrate or run controls you should first adjust the probe
height for the appropriate plate. Refer to "Adjust the Sample Probe
Vertical Height" on Page A-9.

Run System xMAP

Calibrators

To calibrate your system with xMAP calibrators:

1. Vortex the xMAP calibrators and controls containers to ensure

homogeneity.

2. Load a microtiter plate with at least three drops of each: CAL1 in

well A1, CAL2 in well B1, CON1 in well C1, CON2 in well D1
and sheath fluid in well E1 through H1 to wash a total of four
times. You can alternately assign different well locations.

3. Click Eject/Retract from the toolbar. The plate holder ejects.

4. Place the plate on the plate holder.

5. Fill the Luminex XYP reservoir with a solution of 70%

isopropanol.

6. Click Eject/Retract from the toolbar.

7. Click Prime from the Maintenance tab. A confirmation screen

appears. Click OK and wait for the Prime to finish (about 1½
minutes).

8. Click Alcohol Flush from the Maintenance tab. A Confirmation

Screen dialog box appears.

9. Click OK and wait until the alcohol flush completes. The Device

Status section in the status bar changes from yellow to green and
displays “Standby”. This takes about five minutes.

10. Click New CAL Targ. from the Maintenance tab to enter or

confirm the calibration lot numbers. An Update CAL Targets
dialog box appears. See Figure 9.

11. Enter the CAL1 lot number.

12. Enter the expiration date. Click the “Expiration Date” field. A

“Select Calendar” button appears on the right side of the field.

Lasers

must be in the
warmed up state
