3. Click Eject/Retract to place the microtiter plate on the plate

holder.

4. Prepare the microtiter plate with sample for processing.

5. Load the microtiter plate on the plate holder, with well A1 in the

upper-left corner. Be attentive to follow the kit manufacturer’s
directions.

6. Click Eject/Retract again to retract the plate holder.

7. Click Start Plate from the Run Batch tab. The system begins

processing the batch’s template commands and acquiring sample
from the wells as the commands appear on the template.

The system takes a specified volume for testing. A horizontal
progression bar appears on the main screen to show the status of
each sample acquisition. After each sample is analyzed,
approximately 160 µL of sheath fluid is dispelled into each well.

As the system analyzes each well, the well shows either a check
mark in a green well for successful acquisitions or an X mark in a red
well for failed acquisitions. Samples may fail analysis for many
reasons, including lack of sufficient events, cancel command, heater
out of range, or plate cancellation.

The system automatically analyzes samples according to the kit
manufacturer’s template commands.

Once you start processing a batch, you cannot use it to reacquire
data. If you click the Open Batch button and then hit the cancel
button, the system defaults to the last open batch. If you have used a
batch for acquisition before, the system will not display it in the open
batch list.

Pause a Batch

The pause command temporarily interrupts the acquisition process.

For instance, you may need to pause the system during acquisition if
the temperature goes out of the designated range, you need to add
sheath fluid, you need to add a new reagent to your sample group, or
for various other reasons:
