Connecting External Components
Functionality of the Profinia
™
protein purification system can be enhanced by add-on
components. The instrument can be connected to a pH monitor, larger buffer bottles
(standard sizes with Bio-Rad methods are 125 ml and 250 ml), larger sample and fraction
tubes (standard sizes with Bio-Rad methods are 15 ml and 50 ml), and the Profinia cooling
accessory.
11.1 pH Monitor
The optional pH monitor (catalog #620-0411) allows the Profinia system to monitor the pH
of the fluid stream of a protein sample. This pH information is displayed on the instrument’s
screen, stored in the run data record, and can be displayed in real time on the computer
screen if the system is connected to a computer running Profinia software. Run data can
also be exported for later viewing and analysis.
The pH monitor kit (Figure 11.1) includes:
•
Bio-Rad pH probe — calomel pH probe, sealed for maintenance-free operation
•
Profinia pH flow cell — minimum swept volume for best response
•
Tubing assembly — connects the flow cell to the instrument
•
Mounting bracket and screws
•

Additional materials required are standard pH calibration buffers, pH 4.0 and 10.0, available
from laboratory supply companies.
To install the pH flow cell and probe:
1.
Use the power switch to turn the instrument off and unplug the power cord.
2.
Use the handles on the back of the unit to turn it so the back of the unit is facing you.
Important: Use care not to spill any fluids when turning the unit.
3. Disconnect and remove the waste bottle (right side of the instrument).
4. Remove the thumbscrews connecting the rear access panel to the instrument; there
are two thumbscrews at the top of the panel, and two near the center.
