To determine and set optimal gate settings:
1. Create a New Advanced batch with DD gate settings at 0 - 32767
(make sure all other settings are appropriate for your sample).
2. On plate layout choose a single well with the Acquire command
to obtain a single sample.
Note:
The sample is representative of the expected assay
samples. It consists of all bead sets combined, reagents, but no
patient samples. Gate positions are dependent upon buffer
composition. Any changes made to your assay buffer
composition may result in a different optimal gate location.
3. Click and drag the right red line to a position on the right side of
the major peak to eliminate aggregate beads.
4. Click and drag the left red line to a position on the left side of the
major peak to eliminate small particles.
5. Note the numerical gate position indicated in the parenthesis in
the lower left corner of the histogram.
6. Use the numbered gate positions from step 5 in templates and
New Advanced Batches for assays with the same buffer
composition.
Note:
You may want to change the X axis of the Dot Plot for
troubleshooting purposes. We highly recommend using the
default settings in all other instances.
