Column headings:
• No: Well number.
• Well ID: Location of well.
• Sample ID: User-supplied or default name of well.
• Cq (Dye 1): Quantification cycle for Dye 1.
• Conc (Dye 1): Calculated concentration of sample for Dye 1.
• Cq (Dye 2): Quantification cycle for Dye 2.
• Conc (Dye 2): Calculated concentration of sample for Dye 2.
• Ratio: Dye 1 divided by Dye 2.
• Comments: User inputted text.
3.14.7.3 Relative quantification cycles
The relative Cq method allows the user to perform relative quantification without using a standard
curve. Instead, the Cq of an unknown sample is normalized to a known sample e.g. a reference
gene (REF) amplified at the same time from the same sample. For many gene expression studies
it is not necessary to determine the absolute amount of a target as evidence of a relative increase
or decrease in expression compared to a reference sample (calibrator) proves sufficient. The
expression level of the gene of interest in the calibrator sample is defined as 1x, with levels in the
unknowns reported relative to it.
