xMAP Carboxylated Microspheres come in various configurations.
The handling of the product varies with the size of the container and
the fill volume. Avoid excessive agitation of the product to reduce
foaming and surface precipitation. The xMAP microspheres will
settle if left undisturbed. Always ensure that the xMAP microspheres
are homogeneously resuspended prior to dispensing. The uncoupled
xMAP microspheres are not monodispersed and tend to aggregate
until coated. Multiple pipetting from the original container may
affect bead concentrations. Protect the xMAP microspheres from
light at all times. Store xMAP microspheres at 2 to 8°C. Follow the
resuspension instructions based on the size of container in use:
• For 1 mL bottles: Remove from refrigeration and allow
equilibration to room temperature. Place on rotator and gently
rotate for 1-2 minutes or mix by gentle inversion immediately
prior to dispensing.
• For 4 mL and 16 mL bottles: Remove from refrigeration and
allow equilibration to room temperature. For new 4 mL and 16
mL bottles, place on a rotator for a minimum of 15 minutes for
complete resuspension. After using approximately 250 µL from
the bottle, you can reduce the rotator time to 1 to 2 minutes. For
these volumes, vortexing and rocking are not recommended.
Bead Selection
Select beads from the standard 100-region bead map. Refer to
Table 1. You should select beads starting from the upper left, reading
across to the end of a row, and then down to the next row:
Bead Concentration
The concentration of beads in an assay determines system speed. To
analyze a 96-well plate in one hour, use at least 5000 beads per target
