Carefully slacken knurled screw of tool mount and positioning aid a little way. Carefully bring
the capillary holder with the capillary into the focal plane by hand. If necessary, bring the
capillary into the field of view by means of horizontal movements of the joystick (X and Y
axis).

When the capillary is in the field of view, bring it carefully into focus (e.g. at a 4 times zoom
factor).

Tighten knurled screw and positioning aid.
This procedure adjusts the capillary so that approximately 80 % of the field of movement is then
above the current position. This setting can be used for all capillaries of the same length. By
means of the positioning aid (section 4.2.3) the universal capillary holder can be dismantled and
refitted in the tool mount as necessary. This does not affect the alignment.
Other setups can also be used specific to application, without any problems.
5.2.2 Definition of Z limit
To protect the tool and the preparation against damage, and for the automatic Inject function, a
limit must be defined. This is relevant mainly to the application of microinjection into adherent
cells.
Once the limit is defined, the capillary is moved upwards manually above the cells. This plane,
from which the various cells are injected, is termed the search plane.

To define the limit, the capillary is set to the desired
position by lightly pressing in the cell.
