III. TIGHTENING PROCEDURE
1. After inserting guard cartridge, tighten holder by hand
until O-rings make contact with the guard cartridge.
2. While holding the 2-1/16” flats, use a 3/4” wrench to
tighten holder cap until O-rings are fully seated, between
5-9 tick marks.
IV. START-UP RECOMMENDATIONS
a. Background (Avoid Pressure Pikes)
It is possible that a SUDDEN high flow rate start at 50-60 mL/min
may contribute to initial disruption of the prep guard cartridge
bed leading to the peak splitting and shortened lifetimes.
It is recommended to start the flow rate up MORE SLOWLY
(over 5 or 10 minutes) to avoid this issue.
b. Start-Up
1. Purge the pumping system of any buffer-containing mobile
phases and connect the inlet end of the column to the injector
outlet.
2. Flush column with 100% organic mobile phase (methanol or
acetonitrile) by setting the pump flow rate to 0.1 mL/min and
increase the flow rate to 50-60 mL/min over 5 minutes.
3. When the mobile phase is flowing freely from the column
outlet, stop the flow and attach the column outlet to the
detector. This prevents entry of air into the detection system
and gives more rapid baseline equilibration.
4. Gradually increase the flow rate as described in step 2.
5. Once a steady backpressure and baseline have been achieved,
equilibrate the column with a minimum of 10-column volumes
of the mobile phase.
6. For added assurance, perform an efficiency test on the prep
guard cartridge before using it. Waters recommends using
a suitable solute mixture, as found in the “Performance Test
Chromatogram”, to analyze the prep guard cartridge upon
receipt. Determine the number of theoretical plates (N) and
use this value for periodic comparisons.
V. INSTALLING AND REPLACING THE GUARD
CARTRIDGE AND CARTRIDGE HOLDER
Install the cartridge holder onto the HPLC system in front of the
column using the rigid connector. The tubing in the rigid connector is
0.020” or 0.5 mm ID. For all other connections, use the same tubing.
Installing and replacing the guard cartridge:
1. Remove the cartridge holder from the system.
2. Unscrew the holder cap from the holder body.
3. Remove the old guard cartridge, if present.
4. Visually inspect the O-rings for damage or debris.
Replace, if necessary (see Section V).
5. Install the new guard cartridge. The direction of the flow
arrow on the guard cartridge and holder body should point
towards the detector.
6. Re-assemble the holder and tighten according
to instructions above.
7. Attach inlet side of assembled cartridge holder to the tubing
from HPLC injector.
8. Flush with 10 volumes of mobile phase at a low flow rate.
9. Attach outlet side of the cartridge holder to the column
inlet using rigid connector.
10. Check for leaks.
