All standard microplates with 6 to 384 wells (maximum plate height: 23 mm incl. lid) that comply with the
following standards can be measured: ANSI/SLAS 1-2004, ANSI/SLAS 2-2004, ANSI/SLAS 3-2004 and
ANSI/SLAS 4-2004.
Predefined plate definition files are installed at the same time as the operator software installation.
6.4
Optimizing absorbance measurements
6.4.1
Flash settings
Quick measurements with 1 flash (measurement) per well can be performed with all plate types; however,
the measuring quality at low light levels depends on the measuring time during which the absorbance
signal can be received.
6.4.2
Settle time
A settle time can be set before measuring a well. The settle time should always be taken into account for
absorbance measurements. Because of the stop-and-go movement of the plate compartment, the meniscus
of the dispensed liquid may continue to vibrate while the signal is being integrated. This can lead to
fluctuations in the measured values. This effect has been observed in the wells of 96-well plates and larger
wells.
6.4.3
Multiple reads per well
The software enables multiple measurements per well (MRW or multiple reads per well) in the modes for
absorbance, upper florescence and lower florescence.
The functions for several measurements per well can be activated in an absorbance or fluorescence
intensity program by placing a check mark in the "Multiple reads per well" checkbox (see figure below).
A list with suitable microplates can be found in the software instructions.
Increase the number of flashes (measurements) per well until the S/N ratio of the empty wells
stops improving or until the measuring time per well becomes unacceptable.
