Before the start of each measurement, make sure that microtest plate position A1 is correctly inserted. Well
A1 must be in the upper left position.

The maximum permitted load of the plate compartment is 100 g.

The maximum permitted plate height is 23 mm (including lid).

6.2

Settings

The following settings are available independent of the corresponding measuring method.

6.2.1

Temperature control

Some assays require temperature control when they are executed. The device can execute temperature
control within a defined range. The temperature control is carried out across the entire plate in a uniform
manner.

If the microplate was not incubated outside of the device, a temperature adjustment must be performed
before the start of the measurement.

Temperature range: 5 °C above ambient temperature to 42 °C.

6.2.2

Kinetic measurements

A plate can be measured repeatedly at identical intervals. The fluorescence signal can vary significantly
over time, particularly with small sample volumes. Depending on the evaporation rate (which is higher for
corner wells and edge wells than for wells in the center of the plate) the down shift of the sample meniscus
competes with the concentration of the fluorescent components in the remaining sample volume. The
fluorescence intensity also depends on the temperature and processes such as photo bleaching.

6.2.3

Shaking microplates

The device features two shaking modes: linear and orbital. The shaking amplitude can be adjusted in
0.5mm increments from 1 to 6 mm. The frequency depends on the amplitude. The shaking time can be
adjusted from 1 – 999 s.

Position the plate in the "incubation position" to maintain a constant temperature and to
guarantee uniformity across the entire plate.

The temperature may vary slightly if the temperature control function is used during shaking.
