2. If necessary, clarify samples by centrifugation at 8,000 x g
for 20 minutes.
Note: The HPLC or LC/MS/MS method is normally developed before
the SPE method. If possible, select a mobile phase that can elute the
analyte(s) from the Oasis SPE cartridge and also gives the required
chromatography on the analytical column.
b. Prepare the Prospekt-2/Symbiosis System According to the
Operator’s Manual
1. Place three solvents in the high pressure dispenser (HPD) for the
Prospekt-2/Symbiosis system: a) 100% methanol, b) water, and
c) 5% methanol in water.
2. Place the Oasis cartridge tray in the Automatic Cartridge
Exchange module.
3. Place prepared samples in the autosampler.
4. Set up the HPLC system with the mobile phase and analytical
column.
Note: If you encounter problems with your Prospekt-2/Symbiosis
systems, contact your Spark Holland representative.
III. strategIes for method oPtImIzatIon on oasIs
methodoLogIes

The following tips can be employed to optimize on-line SPE methods
in general.
1. Increase the flow rates from solvation, equilibration, and wash to
shorten the process time.
2. Do not exceed the recommended backpressure of 300 bar (4,500 psi).
3. To improve extraction efficiency, reduce the flow rate during the
loading step (depending on the analyte).
4. Use solvent mixes having greater or lesser polarity to produce a
cleaner extract (for example, 60/40:IPA/acetonitrile).
5. For very polar analytes, use 100% pH-modified water for wash
and elution solution.
6. For very non-polar analytes, use stronger solvents, such as
70/30:acetonitrile/methonol.
Note: The achiever better cleanup, optimized method(s) can be used.
