The selection of the best mobile phase will depend on the mix of
analytes and may require some method development on both the
loading/washing steps as well as the elution phase. Reconditioning
of the extraction column is critical for minimising any possible
carryover. It may be found that a re-condition with 100% methanol
is sufficient for this purpose. Complex matrices may require more
stringent re-conditioning, such as a mix of methanol, acetone and
hexane. This re-conditioning step and re-equilibration prior to the
next sample loading are completed during the parallel elution
phase of the second extraction column. Solvents that be used for
re-conditioning are:
• Water
• Methanol
• Isopropanol
• Acetonitrile
• Acetone
• Tetrahydrofuran
• Hexane
