Direct Connect HP 2.1 x 30 mm column makes it possible
to analyze specific analytes in a sample matrix when combined with
Waters ACQUITY UPLC

columns and online SPE technology.

1. INTRODUCTION
The XBridge packing materials were designed to provide excellent
peak shape, high efficiency, and excellent stability for acidic and basic
mobile phases. The recommended operating pH limits for XBridge Direct
Connect HP columns are pH 1-12. For more information on XBridge
HPLC analytical columns, please refer to the XBridge Columns Care and
Use Manual (P/N 720001366EN). The XBridge Direct Connect column
can be screwed directly into the switching valve or connected to fit-
tings like a conventional HPLC column.

II. SAmPlE PREPARATION
Aqueous Samples: Water, Beverages
Adjust pH to maximize analyte retention on the SPE sorbent. Buffer
salts and dispersive agents may be used to increase partitioning onto
the SPE sorbent. Pretreatment to remove suspended matter prior to
SPE treatment may include filtration or centrifugation.
Non-Aqueous Liquid
When appropriate the sample may be diluted with aqueous buffers
and organic co-solvents for reversed-phased SPE. If sufficient dilution
has occurred, the sample may be treated in a manner similar to an
aqueous sample.
III. EQUIPmENT
Shown in Figure 1 is a schematic of Waters UPLC
®
system with online
SPE technology, which consists of a 2777 high-pressure sample
manager, an ACQUITY UPLC
®
quaternary solvent manager (QSM), an
ACQUITY UPLC binary solvent manager (BSM), two extraction columns,
an analytical column, and detection system (mass spectrometry, PDA
or fluorescence). The extraction columns are connected to a switching
valve so they can be operated in parallel.
