microspheres are not monodispersed and tend to aggregate until
coated. Multiple pipetting from the original container may affect
bead concentrations. Protect the microspheres from light at all times.
Follow the resuspension instructions based on the size of container in
use:
For 1 mL containers:
Remove from 2-8°C and allow equilibration to
room temperature. Centrifuge for 1-2 minutes at
≥ 8,000 x g.
Sonicate in a water bath sonicator to disperse the xMAP microsphere
pellet. Mix by gentle vortexing immediately prior to dispensing.
Inversion is not recommended.
For larger containers:
Remove from 2-8°C and allow equilibration to
room temperature. Place on rotator and rotate for a minimum of 15
minutes. Mix by gentle inversion immediately prior to dispensing.
For these containers, vortexing and rocking are not recommended.
Bead Selection
You select beads from the standard 100-region bead map. Refer to
Table 1. Luminex recommends that you select beads in this order,
starting from the upper left, reading across to the end of a row, and
then down to the next row:
Bead Concentration
To a great extent, the concentration of beads in an assay determines
system speed. To analyze a 96-well plate in one hour, we recommend
you use at least 5000 beads per target and do not exceed a bead
concentration of 2.5 x 10

microspheres/mL.
