The appearance of the electropherogram depends on the type of RNA assay performed:

mRNA analyses often yield broad peaks or a smear and, on occasion, contaminating 18S
and 28S rRNA peaks (Figure A.1). In addition, mRNA samples require more time for analysis
than total RNA samples to ensure inclusion of the population of longer transcripts and to
achieve a smooth baseline

Total RNA analyses yield 2–3 prominent peaks corresponding to rRNA (18S and 28S and
occasionally 5S in eukaryotic sources, 16S and 23S in prokaryotic sources, Figure A.2)
