RQI color indicators and value assignments can be customized. The default values are
approximations. Acceptable RNA quality varies depending on the application. Validate your RQI
values for your application and set the ranges accordingly.

Experion software also issues RQI alerts when RQI calculations may not be reliable. When one of the
following limitations or anomalies is detected, an alert is displayed in the Run Summary tab:

RNA conc. too low — concentration of RNA is too low (<10 ng for the RNA StdSens assay
and <200 pg for HighSens assay)
n
Critical anomaly — RQI calculations cannot be performed, sample should be assayed
again (if this error occurs in >1% of samples, contact Bio-Rad Technical Support)
To ensure the best results from RQI analysis, make sure that:
n
Ladder peaks are identified correctly
n
Lower marker is identified correctly
n
18S and 28S fragments are within normal time windows (this may require that you edit a
fragment)
