MAP Technology
reporter
reporter
A molecule (or combination of molecules) with a specific range of
excitation and emission wavelengths that are used to identify or
quantify an analyte. Examples of acceptable reporters for the
Luminex 100 analyzer are Phycoerythrin and Alexa 532.
reporter channel (RP or RP1)
A specific range of wavelengths that includes the emission
wavelength of a designated reporter molecule.
reporter system
Any combination of molecules in solution that serve as reporter for a
particular assay.
RP1
Refers to a reporter molecule bound to the surface of the xMAP
microsphere. Also see reporter channel.
sample
The mixture of assay components (xMAP microspheres, reporter,
patient diluent) that are analyzed.
session
A folder that contains individual assay data and specified settings.
signal
Detectable measurement unit of the reporter molecule.
singlet(s)
A single xMAP microsphere or a population of single xMAP
microspheres. Compare to doublet.
spillover
A circumstance in which one fluorochrome’s emission spectra
overlaps the emission spectra of another fluorochrome generating an
abnormally high intensity measurement. A mathematical technique
called compensation is often applied to correct this occurrence.
suspension
Solution consisting of homogeneously dispersed xMAP
microspheres in an aqueous medium.
template
A folder that contains specific settings and is used to initialize a new
session.
unconjugated
microspheres
An xMAP microsphere set that has been chemically modified so that
its surfaces are covered with some functional group, but that have not
been chemically prepared for use as a component in an assay system.
These base xMAP microspheres do not carry biomolecular reactant.
verification
A process used to ensure the Luminex 100 analyzer is functioning
properly with current calibration settings.
