32. Air dry the plates for about 30 min using
an electric fan. Never oven dry the
plates you are planning to run in further
electrophoresis or chromatography
steps because you may “bake” the
peptides into the cellulose (see fig. AA).

The apparatus can be used to
electrophorese plates sequentially. We
have reused the same buffer up to ten
times without any obvious change in
electrophoresis patterns. Simply wet the
next plate in line and place it on the
apparatus when the previous plate is
finished. (See figure BB).
Note: An alternate method can be used for spotting samples of large volume (10-30
μl) or which
contain large amounts of protein (>20
μg). Spot the sample on a vertical line emanating about
1cm upward from the sample origin mark. Spot 0.5
μl at 1mm increments, drying after each drop,
until the entire line has been spotted once. Repeat this again until the entire sample is loaded.
The plate is wetted using a modified blotter that has a hole cut to the shape of the sample origin
(a long ellipse or rectangle). The sample is electrophoresed and dried as described above.
