B. Initial Preparation of Thin Layer Cellulose Plates 
 
1. 
Select a thin layer cellulose plate. We 
use 100
μm x 20x 20cm cellulose plates
(E.M. Laboratories) (Fig. A). To do this 
first make sure that the cellulose layer is 
of a uniform thickness with an even 
surface that does not have any gouges

Next, place the plate on a light box and 
illuminate from beneath to see if the 
thickness of cellulose is even across the 
area of electrophoresis and 
chromatography (Fig. B). Based on this 
criteria most plates are suitable, 
although we often come across boxes 
(25 plates) that contain several plates 
with an unusable cellulose surface. We 
also recommend using one orientation

 Use the stencil outline in the Appendix 
(Section 6) to make several orientation 
marks on the plate. Place the outline on 
a light box (Fig. C).
