E. Final Preparation of TLC Plate

The plate needs to be wetted with the 
same buffer used in the tanks before it is 
ready for electrophoresis. We use a 
special blotter that wets the plate while 
also concentrating the sample. It is 
composed of two sheets of Whatman 
3MM paper sewn together and has two 
holes about 1.5-2.0cm in diameter cut at 
positions corresponding to the sample 
and dye application marks (use the 
template in the appendix to align the 
holes). Soak the blotter in buffer in a 
large tray (Fig. S).

Let the blotter drain to remove excess 
liquid (Fig. T) then blot it against dry 
paper until it is only slightly damp using

Blotters can be reused until they literally 
fall apart. Used blotters should be 
labeled depending on which buffer they 
have been used for, and rewet only with 
the same buffer. Place the TLC plate on 
a flat surface, cellulose side up, then 
carefully place the damp blotter down on
