29. The plate should be oriented so that the
dye and sample marks are on a vertical 
axis from top to bottom, which should be 
perpendicular to the electric field (edges 
of the wicks). Remember that the anode 
(+red) will be on the left, and the 
cathode (-black) on the right. Most of 
the peptides will migrate toward the 
cathode or to the right during 
electrophoresis if using either pH1.9 or 
4.72 buffers. (PAAs go left). Next, 
place the wicks so they overlap onto the 
cellulose by about 1cm. Cover the plate 
and wicks with the top polyethylene 
sheet, followed by the Teflon pad, then 
the neoprene pad (Fig. X). 
 
 
 
 
 
 
 
 
 
30. Close the lid and again secure the pins.
Next, turn the air pressure up to 10psi 
as before and make sure that the water 
supply is on for cooling the device 
during the run. (See Fig. Y). 
 
 
 
 
 
 
 
 
 
 
 
 
 
 
31. Close the safety cover of the apparatus, 
then set the voltage timer and turn 
mains switch and the power supply on: 
For this run, with pH1.9 buffer, we ran 
for 25min at 1.0kV (
∼21mA (Fig. Z).
After the run is complete, turn the 
power supply off and the mains power, 
then release the air pressure. 
Disassemble the apparatus and 
carefully remove each layer then the
