1. Mount the capillary in the actuator grip head (see Mounting the capillary on p. 14).
2. Using the CellTram vario, fill the mounted capillary with oil from the rear.
3. Absorb the medium via the front capillary opening.
4. Initially, select a higher setting (e.g., 20) for the Int. (intensity) parameter.
Alternative capillary filling
•
Weigh down the capillary by filling it with liquids with higher specific densities.
•
Fill the capillary with a liquid with a high specific density, free of air bubbles, and then with oil.
In this case, reduce all parameters, especially Int. (intensity).
5.4.2
Preparing capillaries with smaller opening diameters
The following section contains a description of how to prepare capillaries with smaller opening
diameters, e.g., for mouse ICSI.
1. Fill the capillary from the rear using a microloader, e.g., Fluorinert™ FC-77.
2. Mount the capillary in the actuator grip head (see Mounting the capillary on p. 14).
3. Absorb the medium via the front capillary opening.
4. Initially, select a lower setting (e.g., 10) for the Int. (intensity) parameter.

Optimizing parameters
The PiezoXpert offers you a wide range of setting options. To determine the optimal parameters
for your application, proceed as follows:
1. Set the parameters for the speed and quantity of the piezo impulses to 1.
Speed: 1
Pulse: 1
2. Set the parameter for Int. (intensity) to a low value (e.g., 10).
Int.: 10 (example)
3. Incrementally increase the value for Int. until the piezo impulses is strong enough to penetrate
the cell wall.
4. Now, fine tune the Speed and Pulse parameters.
Or, you can start with a high value for the intensity (e.g., Int.: 30) and optimize the strength of the
pulse as the values decrease.
If you do not have success with the optimized parameters, you have to take into consideration
that the PiezoXpert cannot deliver the required contribution on its own.
