The time can be set in hours (h), minutes (m) or
seconds (s). Type in the time required followed
by the unit. Use the to move across the
screen to the ramp rate.

The ramp rate is automatically set to the
maximum 3°C per second. A number less than
2°C/s can be inserted. Selecting "Max" will
reset the ramp to the maximum.

Selecting "OK" leaves the Edit program
function menu and returns to the program.

Selecting "Continue" adds another new
step to the program.

If a segment or the Inc/Dec function is inserted
then the software will automatically assign 1
cycle to this stage or loop, as it is referred to in
the software.

INSERTING CYCLE NUMBERS

Selecting "Number Cycles" allows the number
of cycles to be increased up to 99. This function
allows the user to easily program the 2 or 3
step section of the PCR which usually involves
between 20 and 45 repeats of 2 or 3
temperature steps. The program will
automatically repeat in the programmed
sequence each of the temperature steps for the
required number of cycles.

INSERTING INCREMENTAL OR DECREMENTAL TIME OR TEMPERATURE

Under normal circumstances, the Hold Temperature of all steps is constant. However, it is possible to
automatically increase or decrease the temperature of a specified step of a programme. Touchdown
amplification of DNA requires a higher temperature during the initial cycles to ensure that only the
specific piece of DNA is amplified and not non-specific products. A decreasing, increasingly less stringent
annealing temperature is then used for the later cycles of the PCR to increase the yield of the product.
