3.4 Running the Heater/Stirrer

1) Fill reservoir with at least 23 Liters of buffer. (refer to section 3.1 for working buffer volume).

2) Turn Heater/Stirrer “On” (Fig 3-3). Reference section 3.2l Setting the Bath Temperature Set Point (page

15).

a) For DGGE, set at 60ºC.

b) For TGGE, select temperature ramp program, see section 4.3.

c) SSCP may require tank buffer to be below ambient temperature (20-25ºC). (references 13 & 14)

i) Attach external chiller to inlet and outlet of cooling coil (located behind heater/stirrer control).

See page 8.

ii) Set chiller temperature 2-5ºC BELOW desired temperature of tank buffer. Run chiller until tank

buffer reaches temperature.

iii) Set DTSK heater/stirrer temperature at desired running temperature. Heater/stirrer will control

final tank temperature.

NOTE: Autotune heater/stirrer with chiller coolant circulating through cooling coil: Refer to section

3.2r. Confirm pump speed is 12% after autotune.

Preparation/Cleaning of Glass Plate for Gel Casting

Hand wash both plates with a high quality lab detergent followed by a complete rinsing with dH

2

O. Air dry or use a

lint-free tissue. Spray/wipe the chosen inner surfaces of the plate set with 95% ethanol and dry with lint-free tissue.
