Assemble the gradient components. (See Fig 5.)
a. Close all gradient maker valves and place a stir
bar in the mixing chamber, the one with the tube
connector port.
b. Attach one end of a piece of tubing to the outlet of
the gradient maker.
c. Run the other end of the tubing through the peri-
staltic pump and attach it to a tubing connector.
d. Use a second piece of tubing to attach the tubing
connector to the red inlet port at the bottom of
the caster.
