Heated Lid
Use this to set the heated lid temperature (in general approximately
10ºC higher than the hottest block temperature in a program).
Preheat Lid
Set to ON to pre-heat the lid to the set temperature before the thermal
cycling program begins.
Sample Cooling
Set to ON to cool the block to 4ºC while the lid preheats.
Pause
Set to ON to pause at the ambient temperature of the block before the
program starts, unless Sample Cooling has been set to ON. In this case
the unit will pause with the block held at 4ºC. The program will not
commence until the unit is manually un-paused.
Initial denaturation
Set to ON for the required hold time to denature the sample and/or
activate the enzyme before the thermal cycling program begins.
Hot Start
Set to ON to pause at the required set temperature. The program will
not continue until the unit is manually un-paused.
Final Extension
Set to ON for the required hold time to add a final extension step to the
program to ensure complete amplification of the products.
Final Hold
Set to ON to hold the block at the required set temperature at the end of
a program until the user manually stops the unit.
Pause
Set to ON to pause the unit before the program starts. The block will
pause at its current temperature unless Sample Cooling has been set to
ON. In this case the unit will pause with the block held at 4ºC and the
program will not commence until the unit is manually un-paused.
Thermal Cycling
Enter appropriate values in the parameter boxes to create the thermal cycling program. Note: if
Display Invalid Data Warnings has been set to ON in the settings, any invalid values will cause the
box to glow yellow and a warning will be displayed at the bottom of the screen advising on the correct
range.
Stage name
Type in a stage name if required. Up to 20 characters can be used.
Cycles
Enter the number of cycles (repeats) for the stage. The maximum is 99.
Ramp rate
Select the required ramp rate. The range is 0.1ºC/s (minimum) to
5.0ºC/s maximum for heating and 3.0ºC/s for cooling. The value can be
entered to 1 decimal place. Ramp rate will affect the heating or cooling
rate from the previous step to the current step.
