3.3 Gel Casting using external gel caster
1. Level external gel caster using white thumb
screws on base. Place Gel tray into external gel
caster. Ends of gel tray friction fit onto place.
Position the comb into receptacle slot in gel tray.
Add additional comb if desired.
2. Make agarose solution according to manufac-
turer’s instructions.
Use caution when handling
hot agarose solution! Pour molten agarose
solution into gel tray. Allow to cool uncovered
until agarose turn opaque. Stains may be added
during this step. Use according to manufacturer’s
recommendations.
NOTE: (Optional storage) Pre-cast gels can be
stored for up to one month if wrapped in plastic
wrap and stored at 4ºC. Remove combs prior to
storing.
3. Remove comb and position the tray in the unit,
remembering, current travels from negative
(black cathode) towards positive (red anode).
Go to Section 3.4
6. Remove comb.
7. Remove black baffles from their casting
position.
8.Turn baffles 180º (handles facing in) and
place into the baffle storage slots provided
at either end of the unit near both terminals.
